ABSTRACT

Background: Trauma induced by endoscopic surgery may inhibit erythrocyte immune function and activate inflammatory responses, however, there are certain differences between different surgical methods. Aims: To investigate the effect of endoscopic mucosal resection (EMR) on erythrocyte immune function and inflammatory cytokines in patients with gastrointestinal polyps. Methods: A total of 315 patients with gastrointestinal polyps in Meishan People's Hospital from Jan. 2011 to Dec. 2016 were collected and randomly allocated into EMR group (n=160) and control group (n=155), and underwent EMR and routine endoscopic electrocoagulation resection, respectively. The resection rate, postoperative erythrocyte immune function, including peripheral blood red-cell membrane complement 3b receptor rosette (C3bRR), immune complex rosette (ICR), CD58 and CD59, as well as serum inflammatory cytokines, including interleukin-1β (IL-1β), IL-6, IL-8 and IL-12 were compared between the two groups. Results: All gastrointestinal polyps were removed successfully in both groups. Less intraoperative complications were observed in EMR group than in control group (3.1% vs. 20.0%, P<0.05). The postoperative levels of C3bRR, CD58 and CD59 on red-cell membrane in EMR group were significantly higher than those in control group (P<0.05), while the postoperative levels of ICR, and serum IL-1β, IL-6, IL-8 and IL-12 were significantly lower in EMR group than in control group (P<0.05). Conclusions: EMR is effective for treatment of gastrointestinal polyps with lower intraoperative complications. Furthermore, it may protect the erythrocyte immune function and suppress the inflammatory responses caused by surgery.

ABSTRACT

Objective To investigate the change of CD35 expression on neutrophils in the peripheral blood and the relationship between the change and disease activity in patient with myeloperoxidase antineutrophil cytoplasmic antibody-associated vasculitis (MPO-AAV).Methods Forty untreated patients with active MPO-AAV(patient group)and forty healthy volunteers (control group) were enrolled into this study,and Bermingham vasculitis activity score (BVAS) for every patient was recorded.Flow cytometry (FCM) was employed to detect the CD35 and MPO expression on the neurtrophil,and enzyme linked immunosorbent assay (ELISA) was taken to test the levels of autoantibody against MPO-Antineutrophil cytoplasmic antibody (MPO-ANCA),fragment a from the activated complement factor B (Ba) and MPO in peripheral blood from both group.All test results were compared between the 2 groups by t test,Non-parametric test,Spearman correlation analysis.In addition,the relations among the laboratory results and the relationship between BVAS and the laboratory results were analyzed respectively.Results Compared with the control group,the expression level,which was represented as mean flourscence indensity (MFI),of CD35 and neutrophil membrane MPO on peripheral blood neutrophils was significantly increased [(2 014±968) vs (1 454±511),t=3.024,P=0.002 and (709±244) vs (580±158),t=2.806,P<0.01,respectively],and the MPO expression level in neutrophils was significantly lower [(1 525±1 033) vs (3 196±2 126),t=-4.468,P<0.01].Ba and MPO levels in serum of the patient group was significantly higher than that in the control group [37.89(26.17,63.14) μg/L vs 27.99(18.64,46.52) μg/L,Z=-2.521,P=0.012 and 546.16(450.55,729.96) U/L vs 327.93(279.02,365.10) U/L,Z=7.121,P<0.01,respectively].In patient group,the expression level of CD35 had a significant positive relationship with peripheral blood neutrophil count (r=0.573,P<0.01),serum Ba (r=0.433,P=0.005) and BVAS (r=0.368,P=0.020),respectively,whereas,there was a negative correlation between the MPO expressed on the neutrophils and that in the neutrophils (r=-0.458,P=0.003),and a positive relationship between MPO-ANCA and BVAS (r=0.351,P=0.026).Conclusion There is significant increased expression of CD35 on the neutrophil of patient with MPO-AAV,which might protect the neutrophil from destruction by the activated complement alternative pathway,and more neutrophils consequently contribute to the MPO-AAV pathogenesis.Inhibition of CD35 expression might become one of the potential new pathways for the treatment of MPO-AAV.

ABSTRACT

Objective Complement plays an important role in the pathogenesis of myasthenia gravis.The formation of membrane attck complex and the damage of neuromuscular junction are inseparable from the activation of complement.Then to investigate the influencing factors of serum complement C3,C4 in children with myasthenia gravis and investigate the degree of influence of related factors.Methods One hundred sixty seven cases of hospitalized or outpatient myasthenia gravis children from the Department of Neurology in Hunan Children's Hospital were collected,including 33 cases of general MG,134 cases of ocular MG,and 36 cases of normal children as control group.The concentrations of serum C3,C4,IgG,IgM,IgA,IgE were detected by immune compare turbid.The influencing factors of complement C3,C4 were investigated and compared.Results The serum C3 levels were 1.07 ± 0.22 g/L,and the serum C4 levels were(0.17 ± 0.05)g/L in the general MG.The serum C3 levels were (1.01 ± 0.20)g/L,the serum C4 levels were(0.20 ± 0.08)g/L in the ocular MG.There were(1.36 ± 0.28) g/L for C3 levels,(0.25 ± 0.11) g/L for C4 levels in the control group.Compared with the control group,there were significant difference in C3 and C4 between the general MG and ocular MG(P <0.01 or 0.05).The partial correlations coefficients of C3 and course of disease,IgG and C3,C3 and C4 were-0.162,0.135,0.446(P <0.01 or 0.05).The multiple linear regression equations were as followed:C4 =0.420 × C3,C3 =0.655 + 1.148 × C4 + 0.008 × body weight-.005 × course of disease.Using univaruate analysis,the effect factors of C4(F =18.151,P =0.000),body weight(F =6.420,P =0.003),course of disease (F =3.015,P =0.039),age × course of disease × body weight (F =2.997,P =0.042) to C3 were significant (P < 0.05,or P < 0.01).Conclusion The C3 levels are mainly affected by C4,body weight,duration and some interaction effects among several impact factors in children with MG.C4 is mainly affected by C3.

ABSTRACT

Purpose In order to explore whether TNF-α could regulate C3 expression in tubular epithelial cell which induced renal interstitial fibrosis after unilateral ureteral obstruction, while whether TNF-a receptor antagonist etanercept could reduce C3 expression in renal tubular epithelial cell which could alleviate renal interstitial fibrosis. Methods Wide type (WT) and C3 knock out (C3KO) mice were separately established to set up sham group and UUO group. Moreover, WT UUO mice were intraperitoneal injected with different concentrations of etanercept. Observed TNF-a and C3 expression in operated kidney after UUO in WT mice and C3KO mice, the correlation between TNF-a expression in operated kidney and C3 positive renal tubules, degree of operated kidney renal interstitial fibrosis. Observed TNFR, NF-κB p65, C3 expression in kidney tubular epithelial cell cytomembrane and degree of renal interstitial fibrosis after intraperitoneal injected with different concentrations of etanercept. Results Nearly all of kidney tubular epithelial cells cytomembrane were TNFR positive, some of kidney tubular epithelial cells cytomembrane were C3 positive, operated kidney was TNF-a strongly positive in WT and C3KO mice after UUO, and C3 positive renal tubules were strongly correlated with TNF-α positive cells which were infiltrated in renal insterium. Degree of renal interstitial fibrosis in C3KO UUO mice were alleviated when compared to WT UUO mice (P<0.01). Expression of C3, NF-κB p65 in kidney tubular epithelial cells were reduced (P<0.01)and degree of renal interstitial fibrosis were alleviated(P<0.01)after etanercept treated in WT UUO mice. Conclusion After UUO, TNF-a and C3 were both participated in renal interstitial fibrosis. TNF-a regulated renal tubular epithelial cell C3 expression via renal tubule epithelial cell transcription factor NF-κB p65 which was involved in renal interstitial fibrosis, while the TNF-a receptor antagonism etanercept may block the pathway, and degree of renal interstitial fibrosis pathological changes were alleviated at last.

ABSTRACT

Objective To study the patients' clinical characteristics and prognosis when only C3 deposition exists in endocapillary proliferative glomerulonephritis and try to understand deeply the role of C3 in kidney damage deeply. Methods The patients who were diagnosed with endocapillary proliferative glomerulonephritis but only had C3 deposited in immunofluorescence(to avoid false positive,C3≥2 ﹢ was included)were selected from Beijing Children's Hospital Affiliated to Capital Medical University during November 2010 to October 2014. Their clinical manifestations,la-boratory examinations,treatments,prognosis,and pathological changes were analyzed,and literature review was performed. Their clinical characteristics and prognosis were summarized. Results There were 11 patients diagnosed with endocapil-lary proliferative glomerulonephritis which had only C3 deposition(≥2 ﹢ ). Nine of them had onset with acute nephritis syndrome(81. 8% ),and 2 cases presented recurrent paroxysmal gross hematuria(18. 2% ). Seven cases were diagnosed with acute post streptococcal glomerulonephritis(63. 6% ). Eleven cases' clinical manifestations were relatively severe, and the complement C3 was significantly lower than the normal(100. 0% ). Their light microscope showed capillary proli-ferative glomerulonephritis,and the electron microscope showed the immune complexes were deposited in the endothelium,the epithelium or the mesangial area. The patients received corresponding treatment respectively,and all the patients had good prognosis during following up of 7 months up to 39 months. Conclusions Streptococcus infection is a common cause in endocapillary proliferative glomerulonephritis with only C3 deposition. The clinical manifestations of some children are similar to post streptococcal glomerulonephritis but relatively severe. Only deposition of C3 without IgG may be involved in another complement activation mechanism.

ABSTRACT

The present study was designed to evaluate the protective effects of ethanol extracts of Rabdosia japonica var. glaucocalyx (Maxim.) Hara (RJ) on lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice and the possible underlying mechanisms of action. The mice were orally administrated with RJ extract (16, 32 or 64 mg(kg(-1)) daily for consecutive7 days before LPS challenge. The ung specimens and the bronchoalveolar lavage fluid (BALF) were collected for histopathological examinations and biochemical analyses. Pretreatment with RJ significantly enhanced superoxide dismutase (SOD) activity and reduced the wet-to-dry weight (W/D) ratio, the levels of nitric oxide (NO) and protein leakage, and myeloperoxidase (MPO) activity in mice with ALI, in a dose-dependent manner. RJ reduced complement deposition and significantly attenuated LPS-induced ALI by reducing productions of inflammatory mediators, such as tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-1β (IL-1β). The results demonstrated that RJ may attenuate LPS-induced ALI via reducing the production of pro-inflammatory mediators, and reducing complement deposition and radicals.

ABSTRACT

Objective To investigate the effects of erythropoietin (EPO) on complement 3a (C3a)-induced renal tubular epithelial to mesenchymal transition. Methods The HK-2 cells were divided into 6 groups namely control group,EPO group,TGF-β group,TGF-β+EPO group,C3a group and EPO+C3a group.The mRNA and protein expressions of α-SMA,E-cadherin and C3 were investigated by RT-PCR,Western blot and immunofluorescence respectively. Results Compared with control group and EPO group,the mRNA and protein expressions of α-SMA in HK-2 cells were up-regulated after the intervention of C3a or TGF-β (all P<0.05).On the contrast,the mRNA and protein expressions of E-cadherin were down-regulated(P<0.05),the mRNA and protein expressions of C3 were enhanced (all P<0.05).However,all those above effects of C3a or TGF-β were inhibited after the intervention of EPO (all P<0.05). Conclusion EPO is capable of suppressing the epithelial to mesenchymal transition induced by C3a.

ABSTRACT

ObjectiveTo investigate the association of complement C3 with clinical and serological features of patients with systemic lupus erythematosus.MethodsData was collected by the same methods in the past ten years in fifteen hospitals in Jiangsu Province and then data weres summarized for retrospective analysis.Clinical and laboratory data were selected and then analyzed by Chi-square test,Wilcoxon rank sum test and Logistic regression.ResultsOne thousand four hundred and five patients were investigated.One thousand and forty two had low serum complement C3 level.In this case control study,hospitalization age,disease course,admission times,pleurisy,gastrointestinal involvement,general lymphadenopathy/hepatosplenomegaly,white blood cell count, haemoglobin level,platelet count, serum C-reactive protein level,serum albumin level,serum creatinine level,Urine protein quantification,anti-nuclear antibodies (ANA),anti-dsDNAantibodies, anti-SmantibodiesandSLEDAIscore were possible factors associatedwith complement C3 reduction(P<0.05).Logistic regression analysis showed that CRP (OR=0.396,0.254-0.617,P=0.000),ANA (OR=2.907,1.267-6.670,P=0.012),urine protein level(OR=1.702,1.043-2.779,P=0.033) and SLEDAI score (OR-0.930, 0.886-0.975,P-0.003) were correlated with complement C3 reduction.Conclusion Complement C3 level is valuable for lupus flare assessment.The complement C3 reduction is a risk factor for renal impairment.

ABSTRACT

OBJECTIVE: To investigate the effects of erythropoietin (EPO) on renal fibrosis of unilateral uretreal obstruction (UUO) rats and its mechanism. METHODS: The model of renal fibrosis was established by UUO procedure in rats. Experimental rats were randomly divided into 4 groups: sham operation group (control group), UUO group, 100 u · kg-1 · d-1 EPO treatment group (low dosage group) and 1000 u · kg-1 · d-1 EPO treatment group (high dosage group). The rats in control group and UUO group were given physiological saline, and those in UUO treatment group were given EPO by intraperitoneal injection from 3 to 14 d after establishment of UUO model. All rats were given methane to induce anesthesia and consequently executed. The pathological change of renal tissue was observed, and immunohistochemistry was performed to measure the expressions of α-SMA, E-cadherin and complement 3 (C3) in renal tissue. RESULTS: Compared with UUO group, renal fibrosis were attenuated by EPO treatment, especially in high dosage group. Compared with control group, the expressions of α-SMA and C3 were significantly up-regulated, and the expressions of E-cadherin were significantly down-regulated in UUO group. In contrast, EPO treatment significantly attenuated the up-regula-tion of C3 and α-SMA and the down-regulation of E-cadherin in renal tissue, and these changes were more obvious in UUO rats treated with high dosage of EPO. CONCLUSION: EPO can inhibit EMT and attenuate renal fibrosis in obstructed kidney. It is suggested that the renoprotective effects of EPO are probably related with inhibition of C3 expression in renal tubular epithelial cells. ABSTRACT

Objective To study the clinicopathological features of C3 glomerulonephritis and the associations between plasma complement fragments level and clinical manifestations.Methods The clinical and pathological data of the 12 patients with C3 glomerulonephritis in our division from January 1999 to June 2010 were analyzed retrospectively.Concentrations of plasma factor B,Ba,C3,C3a,C4a and C5a were detected by commercial available sandwich ELISA kits on the day of renal biopsy.Results Ten of the 12 patients were C3 glomerulonephritis without MPGN,and the rest 2 were C3 glomerulonephritis with MPGN.All the patients presented proteinuria.Two of the 10 C3 glomerulonephritis patients without MPGN presented nephrotic range proteinuria,6 with microhematuria,1 with gross hematuria,and 2 with renal insufficiency.One of the 2 C3 glomerulonephritis patients with MPGN presented nephrotic range proteinuria,accompanied by microhematuria,hypertension and renal insufficiency.The other patient showed moderate proteinuria with normal renal function.Most of C3 glomerulonephritis patients without MPGN showed mild mesangial proliferative glomerulonephritis,and 4/10 patients had various degree cresentic formation.One C3 glomerulonephritis patient with MPGN had 47.1% cresentic formation.The concentration of plasma C3 in C3 glomerulonephritis patients was normal,while the plasma factor B was significantly decreased,and the concentrations of plasma Ba,C3a,C4a and C5a were significantly elevated.The concentration of plasma Ba was positively correlated with the proteinuria level,while the concentrations of plasma C3a,C4a and C5a were not correlated with the levels of proteinuria or plasma creatinine.Conclusions Majority of these 12 patients were C3 glomerulonephritis without MPGN,manifests as nephritic syndrome clinically and mild mesangial proliferative glomerulonephritis histopathologically.Complement activation via alternative pathway may play an important role in the pathogenesis of C3 glomerulonephritis.

ABSTRACT

Objective:To investigate the influence of B-lymphocyte chemoattractant on the immune response of CVB3 fusion gene vaccine pcDNA3/C3d3-sVP1.Methods:BALB/c mice were divided into 4 groups randomly, and injected intramuscularly with pcDNA3,pcDNA3/BLC,pcDNA3/C3d3-sVP1 and the combination with the plasmid pcDNA3/BLC and pcDNA3/C3d3-sVP1.At a certain time,they were measured for the titers for neutralizing antibodies,specific CTL cytotoxic activity.The protective efficacy of DNA vaccinations was evaluated by titers of blood viruses and survival rate.Results:The titers for antibodies increased with the time of inoculation.More specifically,the antibody titers (42.17±1.43) and the specific CTL cytotoxic activity (41.3%±3.51%) of the mice in the combination group were remarkably stronger than in the mice with pcDNA3/C3d3-sVP1(P<0.05),but the virus titers of blood was lower.After lethal CVB3 challenge,the protection of mice from death in the combination group with the plasmid pcDNA3/BLC and pcDNA3/C3d3-sVP1 was 44%.Survival curves indicated that the survive state of combination group was better than others.Conclusion:BLC can strongly enhance the specific immunity induced by C3d3-sVP1.

ABSTRACT

Objective To explore the value of serum SC5b-9, anti-C1q antibody, C3 and C4 levels in the assessment of lupus activity. Methods The enzyme linked immunosorbent assay (ELISA) was used to measure SC5b-9 and anti-C1q antibody, rate nepheiometry was used to detect the serum level of C3 and C4 in sera of 62 SLE patients, 35 patients with other rheumatic diseases (including rheumatoid arthritis, ankylosing spondylitis, primary Sjogren' s syndrome, mixed connective tissue disease, dermatomyositis, polymyositis, systemic sclerosis and vasculitis) and 35 healthy controls. And the correlation between above-mentioned parameters and lupus clinical manifestations, disease activity and histological type of lupus nephritis were analyzed. Results In SLE patients, the levels of SC5b-9 and anti-C1q antibody were significantly higher than those in patients with other rheumatic diseases and healthy controls (P<0.05). The titers of SC5b-9 and anti-C1q antibody negatively correlated with C3 and C4 (P<0.05), and positively correlated with SLEDAI (P<0.05). The sensitivity and specificity of the combination of these three measurements for SLE was 95.37% and 98.46 respectively. SC5b-9 and anti-C1q antibody were associated with the presence of proliferative glomerulonephritis (P <0.05). Conclusion Taking the evaluation of all these three measurements simultaneously is valuable for the diagnosis of lupus flare. SC5b-9 and anti-C1q antibody may play major roles in the immunopathogenesis of lupus nephritis.

ABSTRACT

A 72-year-old woman presented with generalized edema and proteinuria. Renal biopsy disclosed highly organized fibrillary deposits in subendothelial area by electron microscopy. The microfibrils were 14 nm in diameter and randomly arranged. They did not have a microtubular appearance. These materials were negative for Congo red staining. Cryoglobulinemia or paraproteinemia including light chains was not found. So we can diagnose her as fibrillary glomerulonephritis (GN). In fibrillary GN serum complement levels are usually normal except in rare cases with systemic disease. Here we present a rare case of fibrillary GN with unusual hypocomplementemia.

ABSTRACT

@#ObjectiveTo investigate the relationship between complement-3(C3) and progression of early cerebral infarction.Methods168 consecutive patients with acute cerebral infarction within the first 24 hours from onset were recruited.Fasting venous blood sample was drawn on the morning of the second or the third day after admission.C3 and other clinical chemistry variables were measured.The relationship of C3 and progressing stroke(PS) was analyzed by Logistic regression.Results68 cases(40.48%) were considered as PS.Level of serum C3 in PS patients was 1.28±0.20 g/L higher than in those with non-PS(1.16±0.25 g/L,P=(0.000)).A higher C3 accompanied a higher tendency of prevalence of PS(P=0.001).Logistic regression analysis showed that C3 was significantly correlated with PS(OR for each 1.0 g/L=7.27;95% CI=1.52~36.39; P=0.013).ConclusionElevated serum C3 may play an important role in the prevalence of PS.

Objective To study complement receptor typeⅠ(CR1)activity of erythrocytes in pa-tients with SLE.Methods Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),CR1rosette(RBC-CR1R)and immuno-complex rosette(RBC-ICR)on red cell,the ery-throcyte complement receptor I type(ECR1)genomic density polymorphism(HH type,HL type,LL type)and erythrocyte CR1immune activity were determined in32patients with SLE and in48normal individuals.Results It was found that HH type rate of ECR1density polymorphism in patients with active SLE was significantly lower(10/16,62.5%)than that(13/16,81.3%)in patients with stable SLE.The level of CR1immune activity in HH type was significantly higher than that in HL,LL type of SLE,and significantly dif-ferent from that in48normal individuals(P

ABSTRACT

Objective To examine the change of erythrocyte immune adherence parameters during the haemodialysis and the influence of different biocompatible dialysis membranes on it.Methods We have observed the dynamic trend of erythrocyte immune adherence parameters at different hemodialysis phase by way of different biocompatible membrane.Results RBC C\-3b declined and RBC IC increased obviously in uremia patients.During the haemodialysis,RBC C\-3b declined even more in the early phase and then increased gradually;RBC IC increased gradually in early phase,and then kept steady.Cu membrane influenced the erythrocyte immune adherence function greatly,Ps membrane moderately,and PMMA membrane slightly.Conclusion The dynamic trends of erythrocyte immune adherence parameters change with the different biocompatible membrances used during hemodialysis.

ABSTRACT

Objective To investigate the characteristic changes in the mRNA and protein of complement 3 in spinal dorsal horn of rats with neuropathic pain,and the role of abnormal activation of complement protein in the mechanism of pain production.Methods Eighty-four healthy male SD rats were divided randomly into seven groups(n=12 for each group):normal control group,sham-operation 1d,3d and 7d groups,and chronic constriction injury of the sciatic nerve(CCI)1d,3d and 7d groups.The left sciatic nerve was ligated loosely in CCI groups,while it was only exposed but not ligated in rats in sham-operation groups.The mechanical and thermal pain thresholds were measured on 1,3 and 7 days after operation,and the mRNA and protein of complement 3 in the dosal horn of the spinal cord were determined respectively by RT-PCR,immunoturbidimetry and immunohistochemistry.Results The mechanical and thermal pain thresholds were observed to be lowered in rats one day after the sciatic nerve ligation,and a state of hyperalgesia was found to be persistent up to 7 days after CCI.This symptom was not observed in sham operation group.The expression of mRNA and protein of complement 3 in spinal dorsal horn were increased on 1,3 and 7 days after CCI.Interestingly,a high expression of mRNA of complement 3 was also observed in rats one day after sham-operation.Both mRNA and protein of complement 3 were not obviously elevated in rats of sham operation 3d,7d groups and normal control group.Conclusion The mRNA and protein of complement 3 in spinal dorsal horn are highly up-regulated in rats with neuropathic pain,suggesting that the characteristic dynamic changes in complement may contribute to the establishment and maintenance of hyperalgesia.

ABSTRACT

The purpose of this study was to determine biochemical parameters related to protein and immunity. Subjects were 125 preschool children(M:69, F:56) residing in low income area of Seoul. Mean serum total protein of the children aged 6 was 7.3g/dl which was significantly higher than 6.6g/dl of the group aged 3. The mean serum albumin was 4.7g/dl for 3, 4, 5 age group, and 4.9g/dl for 6 age group and there was no significant difference. Serum retinol binding protein(RBP) is used as a sensitive indicatior of protein, becase it tends to fall rapidly in response to protein status and respond to quickly dietary treatment. Mean RBP for each group(3, 4, 5 and 6 age group) were 2.5 microgram/dl was 15.9%, 19.2%, 24.3% and 16.7%, respectively. The 24-hour urinary excretion of hydroxyproline was 7.9 mg, 14.6 mg, 11.7 mg and 11.8 mg for each group and the mean excretion of all children was 12.2 mg/day. Children aged 3 were excreting significantly lower amount of hydroxprolinc per day than the children aged 4. The mean hydroxyproline index were 2.18, 2.39, 2.52, 2.80 for each age group and the mean of a group aged 6 was significantly higher than that of the group aged 3. The proportion of children assessed as malnourished and impaired growth(hydroxyproline index<2.0) was 18.8%, 4.9%, 2.5% and 4.3%, respectively. The nutrients which showed significant relationship with protein and immunity parameters were niacin, vitamin C and calcium. Vitamin C showed significant positive relationship(p<0.05-p<0.01) with serum RBP, total protein and globulin. The triceps skinfold thickness was significantly and positively correlated with serum globulin. Serum IgG showed significant positive relationship with height, weight, girth of chest and midarm circumference(p<0.05-p<0.01).

ABSTRACT

Higher levels of IgE-containing immune complexes (IC) have been reported in sera from patients with allergic diseases than in sera from controls. To evaluate the possibility of an IC-mediated mechanism in the pathogenesis of bronchial asthma, we measured circulating C3-containing IgE IC (C3-IgE IC) using anti-C3 ELISA from 20 house dust mite (HDM)-sensitive asthmatics, 20 non-atopic asthmatics, and 14 non-atopic controls. C3-IgE IC levels were significantly higher in HDM-sensitive asthmatics (mean +/- S.D.: 12.2 +/- 7.8 AU/ml) than in non-atopic asthmatics (6.5 +/- 7.5 AU/ml) or controls (5.8 +/- 4.4 AU/ml). C3-IgE IC levels were significantly correlated with HDM-specific IgE levels (r = 0.50, p<0.05), but not with total IgE levels (r = 0.36, p< 0.05) in HDM-sensitive atopic asthmatics. C3-IgE IC levels in sera did not significantly change during HDM-bronchoprovocation test in six HDM-sensitive asthmatics who showed positive reaction. Part of C3-IgE IC could be precipitated by protein G coupled beads. In conclusion, C3-IgE IC levels were elevated in sera from HDM-sensitive asthmatics; moreover IgG antibodies might be a component of C3-IgE IC. Our results suggest that an IgE IC-mediated mechanism could be involved in the pathogenesis of atopic asthma.
